Mark, Apeks DST first stage (which is the usual first stage supplied with
TX40 and TX50 second stages here in the UK) has the three 3/8 and one 1/2
LP ports.

The 1/2 LP port is intended to be used for your primary second stage, and
so would always have a hose on it, the correct hose size should have been
supplied with your primary.  What I don't know is if this has a real
performance benefit, or if it is a gentle scam to get you to buy Apeks LP
hoses...

I have a follow up question on this, (for anyone else who uses Apeks -
Mike?)

My primary TX40 is on a 1.5m (5 foot) hose, and the backup TX40 on an
approx. 50cm/20 inch necklaced hose.  While the long hose wraps snugly for
open water use, the backup hose sticks up and out a little more than I
would like due to the backup's LP port pointing slightly back from the
valve.  If I got a long hose for the 3/8 port and put my primary on that,
and have the backup on a short hose from the 1/2, this would improve hose
routing from the first stage, and make the backup hang more comfortably.
Any reason why I shouldn't do this?
