Absolutely not ... actually a HOSTS file is beneficial if DNS is not used.
 LMHOSTS can also be used as well.  This sounds like on of my earlier
posts on the 5.0 problem - and if there was a misunderstanding - please
accept my apologies...

They can be on different routed segments, however, the subnet for the 3000
needs to be placed into the smb.conf file after the IP address.  DNS
and/or your routers should also help in this regard.
